Research Excerpts

Overview

2'-deoxyguanosine is a key medicinal intermediate that could be used to synthesize anti-cancer drug and biomarker in type 2 diabetes. 8-oxodeoxyguanusine (8-oxodG) is a highly mutagenic oxidative DNA lesion that is formed by the oxidation of deoxyGuanosine.

Toxicity

2'-deoxyguanosine (dGuo) is toxic to normal T and B lymphocytes, an effect mediated by intracellular accumulation of guanine ribonucleotides. PNP deficiency is a severe cellular immune defect of inherited purine nucleoside phosphorylase (PNP) deficiency.

Dosage Studied

The time courses of lipid peroxidation, free radical generation, and 8-hydroxydeoxyguanosine (8OHdG) formation were used to assess the level of oxidative stress in the liver of B6C3F1 mice dosed orally once daily, 5 days a week for 8 weeks. Dose-response studies in which decreasing doses of the carcinogen resulted in fewer numbers of adducts showed that significantly higher levels of 8-Hydroxyde Oxyguanosines could be detected in liver.
