Description

Hycanthone is a synthetic antischistosomal drug. It is a thioxanthone derivative that acts as a DNA intercalator, inhibiting DNA replication and transcription. Hycanthone was synthesized in the 1960s and was the first effective treatment for schistosomiasis, a parasitic disease caused by Schistosoma worms. The drug is known to be effective against Schistosoma mansoni, the parasite responsible for most cases of schistosomiasis in Africa and South America. Hycanthone is administered orally or intravenously. However, it has several significant side effects including hematologic, gastrointestinal, and neurological effects. The drug is also known to be mutagenic and carcinogenic, which has limited its use in many areas. Due to its toxicity and the development of newer, safer antischistosomal drugs, hycanthone is no longer widely used. However, it remains important for studying the mechanisms of drug action and for developing new drugs to treat schistosomiasis.'

Hycanthone: Potentially toxic, but effective antischistosomal agent, it is a metabolite of LUCANTHONE. [Medical Subject Headings (MeSH), National Library of Medicine, extracted Dec-2023]

hycanthone : A thioxanthen-9-one compound having a hydroxymethyl substituent at the 1-position and a 2-[(diethylamino)ethyl]amino substituent at the 4-position. It was formerly used (particularly as the monomethanesulfonic acid salt) as a schistosomicide for individual or mass treatement of infection with Schistosoma haematobium and S. mansoni, but due to its toxicity and concern about possible carcinogenicity, it has been replaced by other drugs such as praziquantel. Dosage Studied

The effects of Astiban, Lucanthone, Hycanthone and Niridazole on autophagic activities in the gastrodermis of Schistosoma mansoni were determined in vivo. A dosage schedule of hy canthone which was too small to have any significant chemotherapeutic effect in mice was sufficient to induce a statistically highly significant incidence of micronodular lesions and of precancerous nodules.
