Valdecoxib is a nonsteroidal anti-inflammatory drug (NSAID) that selectively inhibits cyclooxygenase-2 (COX-2). It was developed by Pfizer and marketed under the brand name Bextra. Valdecoxib was approved by the FDA in 2001 for the treatment of acute pain, dysmenorrhea, and osteoarthritis. It was also approved for the prevention of colorectal cancer in patients with familial adenomatous polyposis. However, valdecoxib was withdrawn from the market in 2005 due to concerns about its cardiovascular safety. Valdecoxib was synthesized by Pfizer scientists in the 1990s. It is a potent COX-2 inhibitor with a long duration of action. Valdecoxib is thought to be effective in treating pain and inflammation because it selectively inhibits COX-2, an enzyme that produces prostaglandins, which are involved in pain and inflammation. Valdecoxib is a prodrug that can be administered intravenously or intramuscularly (parecoxib, Dynastat) and has been developed for the short-term treatment of postsurgical pain. It has a tolerability profile superior to that of oxycodone/acetaminophen.

Valdecoxib has been entrapped in sub-5 microm particles of poly[Lac(Glc-Leu)] copolymer using a oil-in-water (o/w) emulsification-solvent evaporation technique. It is almost superimposable at the site critical for COX-2 inhibition, a structural side pocket in the enzyme which coincides with the sulfonamide group of the drug.
