Description

Methiothepin is a synthetic, non-selective serotonin antagonist that binds to serotonin receptors, particularly 5-HT2A receptors. It was initially developed in the 1960s as a potential antipsychotic, but its clinical use was limited due to side effects like sedation and orthostatic hypotension. Methiothepin is primarily used in research to study the role of serotonin in various physiological and pathological processes, including sleep, mood, anxiety, and neuroprotection. It is also used to investigate the mechanisms of action of other serotonin-related drugs.'

Methiothepin: A serotonin receptor antagonist in the CENTRAL NERVOUS SYSTEM used as an antipsychotic. [Medical Subject Headings (MeSH), National Library of Medicine, extracted Dec-2023]

methiothepin : A dibenzothiepine that is 10,11-dihydrodibenzo[b,f]thiepine bearing additional methylthio and 4-methylpiperazin-1-yl substituents at positions 8 and 10 respectively. Potent 5-HT2 antagonist, also active as 5-HT1 antagonist. Differentiates 5-HT1D sub-types. Also displays affinity for rodent 5-HT5B, 5-HT5A, 5-HT7 and 5-HT6 receptors (pK1 values are 6.6, 7.0, 8.4 and 8.7 respectively). Research Excerpts

Effects

Methiothepin at 0.1 microM has been shown to shift to the right the concentration-effect curve of D-lysergic acid diethylamide (LSD) on stimulation-evoked release of [3H]5-HT.

ExcerptReferenceRelevance
"Methiothepin has been reported to induce an increase of specific binding sites for 3H-5TH 2-3 days following a single administration of a large dose. "( Reduced head-twitch response to quipazine of rats previously treated with methiothepin: possible involvement of dopaminergic system.

Actions

Methiothepin did not inhibit pulmonary vascular contractility per se, because the pulmonary hypertensive response to the thromboxane A2 mimetic U44069 remained intact. Methiot hepin produced an increase in arterial blood pressure when injected into the left side.

Treatment

Methiothepin-pretreated rats exhibited a significantly reduced response after quipazine but not a significant change after L-5HTP. Pretreatment with methiot hepin and mesulergine, but not clozapine, inhibited (R)-8-OH-DPAT-induced monosynaptic reflex depression.
