Description

Eticlopride is a dopamine D3 receptor antagonist with high affinity for the D3 receptor, but also moderate affinity for the D2 receptor. It is used in research to study the role of dopamine in various physiological and pathological processes. The synthesis of eticlopride involves a multi-step reaction sequence starting with the preparation of the benzamide derivative and subsequent modification to incorporate the desired pharmacophore. Eticlopride has been shown to have antipsychotic-like effects in preclinical models and has been investigated for its potential therapeutic use in schizophrenia and other neuropsychiatric disorders. The compound has been studied extensively for its potential to improve cognitive function and reduce negative symptoms in schizophrenia. However, eticlopride has not yet been approved for clinical use, and further research is required to evaluate its safety and efficacy in human subjects. Eticlopride is a valuable research tool that has contributed significantly to our understanding of the role of dopamine in the brain and its potential therapeutic implications.'

Overview

Eticlopride is a substituted benzamide analog with high affinity and selectivity for dopamine (DA) D2-like receptors. It was initially developed as a potential antipsychotic agent.

"The eticlopride-induced increase in DARPP-32 phosphorylation was also decreased by pretreatment with the D(1) receptor antagonist SCH 23390 (0.125 and 0.25 mg/kg) and completely reversed by combined pretreatment with SCH 23390 (0.25 mg/kg) plus SCH 58261 (10 mg/kg)."( Regulation of the phosphorylation of the dopamine- and cAMP-regulated phosphoprotein of 32 kDa in vivo by dopamine D1, dopamine D2, and adenosine A2A receptors.

Treatment

Eticlopride pretreatment also attenuated METH-induced DAT monomer loss and decreases in DAT function. The attenuation was much less pronounced than the effect on DAT complex formation. Pretreatment had no effect on the dopamine-induced rate of [3H]SCH 23390 dissociation.
