Description

Triparanol, also known as MER-29, is a synthetic compound that was developed in the 1950s as a cholesterol-lowering drug. It works by inhibiting the enzyme delta-24-reductase, which is responsible for converting desmosterol to cholesterol in the biosynthesis pathway. Triparanol's mechanism of action was initially believed to be promising, as it effectively reduced cholesterol levels in patients. However, the drug was withdrawn from the market in 1962 due to serious adverse effects, including cataracts, peripheral neuropathy, and liver damage. Despite its withdrawal, triparanol continues to be studied for its potential therapeutic effects in other conditions, such as cancer and Alzheimer's disease. Current research focuses on understanding the mechanism of action of triparanol and its potential for drug repurposing.'

Triparanol: Antilipemic agent with high ophthalmic toxicity. According to Merck Index, 11th ed, the compound was withdrawn from the market in 1962 because of its association with the formation of irreversible cataracts.
