The series of molecular signals initiated by binding of a Wnt protein to a frizzled family receptor on the surface of a target cell and ending with a change in cell state. [PMID:11532397]
The Wnt signaling pathway is a fundamental cell-cell communication system that plays a crucial role in various developmental processes, from embryonic patterning to adult tissue homeostasis. It is a highly conserved pathway across different species and involves a complex interplay of secreted proteins, cell surface receptors, intracellular signaling molecules, and transcription factors. The Wnt signaling pathway is initiated by the binding of Wnt proteins, a family of secreted lipid-modified glycoproteins, to their cognate receptors, Frizzled (Fz) proteins, located on the cell surface. Upon Wnt binding, Fz receptors activate Dishevelled (Dvl), a cytoplasmic protein that acts as a central hub in the pathway. Dvl then activates a downstream signaling cascade that can proceed through two major branches: the canonical Wnt pathway (also known as the β-catenin pathway) and the non-canonical Wnt pathway.
The canonical Wnt pathway is characterized by the accumulation of β-catenin, a key transcription factor, in the cytoplasm. In the absence of Wnt signaling, β-catenin is continuously phosphorylated by a protein complex consisting of Axin, adenomatous polyposis coli (APC), glycogen synthase kinase 3 beta (GSK3β), and casein kinase 1α (CK1α). This phosphorylation targets β-catenin for degradation by the proteasome. However, when Wnt binds to Fz, Dvl activates the downstream signaling cascade, leading to the inhibition of the β-catenin degradation complex. This allows β-catenin to accumulate in the cytoplasm and translocate to the nucleus, where it associates with T-cell factor/lymphoid enhancer factor (TCF/LEF) transcription factors to activate gene expression.
The non-canonical Wnt pathway, also known as the planar cell polarity (PCP) pathway, regulates cell polarity and migration, as well as cytoskeletal reorganization. This pathway involves different downstream signaling molecules, such as Rho GTPases and JNK kinases, and does not directly involve β-catenin.
