Description

Decamethonium is a synthetic neuromuscular blocking agent that acts as a competitive antagonist at the nicotinic acetylcholine receptor at the neuromuscular junction. It was initially developed in the 1950s and was one of the first muscle relaxants to be used in clinical practice. Decamethonium produces a long-lasting blockade of neuromuscular transmission, leading to muscle paralysis. Its mechanism of action involves binding to the acetylcholine receptor, preventing acetylcholine from activating it and triggering muscle contraction. The compound was synthesized from a reaction of bis(2-chloroethyl)amine with hexamethylenediamine. Due to its long duration of action and potential for cardiovascular side effects, decamethonium has largely been replaced by other neuromuscular blocking agents with more favorable pharmacokinetic profiles. However, it remains of interest in research studies investigating the structure-activity relationships of neuromuscular blocking drugs and the development of novel therapies for various neuromuscular disorders. Its importance lies in understanding the fundamental mechanisms of neuromuscular transmission and the development of pharmacological tools for studying muscle function. The long-lasting blockade of neuromuscular transmission caused by decamethonium makes it a valuable tool for research studies investigating the effects of muscle paralysis on various physiological processes, including breathing, circulation, and metabolism.'

Dosage Studied

Decamethonium bromide was administered to a series of embryos in five dosage groups, producing a cohort of embryos with various degrees of paralysis and atrophy of tendons and muscle. To assess whether this interaction is also observed with the depolarizing drug, decameth onium, the potency of decam ethonium and vecuronium was assessed using a cumulative dose-response technique.

Roles (2)

RoleDescription
muscle relaxantA drug used to produce muscle relaxation (excepting neuromuscular blocking agents). Its primary clinical and therapeutic use is the treatment of muscle spasm and immobility associated with strains, sprains, and injuries of the back and, to a lesser degree, injuries to the neck. Also used for the treatment of a variety of clinical conditions that have in common only the presence of skeletal muscle hyperactivity, for example, the muscle spasms that can occur in multiple sclerosis.
nicotinic acetylcholine receptor agonistAn agonist that selectively binds to and activates a nicotinic acetylcholine receptor.
