Cellular response to copper ion is a complex process involving a series of tightly regulated steps that ensure proper copper homeostasis. The process begins with the uptake of copper ions from the extracellular environment. Copper transporters, such as CTR1, facilitate the entry of copper into the cytoplasm. Once inside the cell, copper is bound to chaperone proteins, such as ATOX1, which deliver it to specific cellular compartments or enzymes.

To prevent copper overload, cells possess efflux mechanisms. ATP7A and ATP7B, transmembrane P-type ATPases, transport excess copper from the cytoplasm to the trans-Golgi network or to the bile canaliculi, respectively. These proteins are crucial for maintaining copper homeostasis and preventing its accumulation to toxic levels.
