Inclusion bodies are protein aggregates that form within cells, often as a result of overexpression or misfolding of recombinant proteins. These aggregates are typically insoluble and can accumulate in various cellular compartments, including the cytoplasm, endoplasmic reticulum (ER), and perinuclear region. The cellular component of an inclusion body is a complex structure that involves multiple cellular components and interactions.

**1. Misfolded Proteins:** The core of the inclusion body is composed of misfolded proteins. These proteins have lost their native conformation and are unable to fold properly. This misfolding can be caused by various factors, including mutations, changes in cellular environment, or the overexpression of the protein.

**2. Chaperones:** Chaperones are proteins that assist in protein folding and prevent aggregation. However, in the case of inclusion bodies, chaperones are often overwhelmed by the large number of misfolded proteins. Some chaperones can become trapped within the inclusion body, contributing to its structure.

**3. Molecular Chaperones:** These are small proteins that act as "folding helpers" to aid in the proper folding of proteins. They can help misfolded proteins refold, but in cases of massive misfolding, they can become trapped in the inclusion body, further contributing to its formation.

**4. Ubiquitin:** This small protein attaches to misfolded proteins, marking them for degradation by proteasomes. However, the proteasome system can be overwhelmed by the accumulation of misfolded proteins, leading to the formation of inclusion bodies.

**5. ER Stress Response:** If the accumulation of misfolded proteins occurs in the ER, it triggers the unfolded protein response (UPR). The UPR is a cellular stress response that attempts to restore ER homeostasis. However, prolonged UPR can lead to cell death.

**6. Membrane Components:** The inclusion body can be surrounded by a membrane that originates from the ER or other cellular membranes. This membrane can contain various proteins and lipids involved in cellular processes, and it can act as a barrier separating the inclusion body from the surrounding cytoplasm.

**7. Cytoskeleton:** The cytoskeleton plays a role in the formation and maintenance of inclusion bodies. Microtubules and intermediate filaments can interact with the inclusion body and influence its location and movement within the cell.

The precise composition and structure of inclusion bodies can vary depending on the protein involved, the cell type, and other factors. However, the general principle is that they represent a complex accumulation of misfolded proteins, chaperones, and other cellular components that are unable to be properly processed and degraded.

Retinaldehyde: A diterpene derived from the carotenoid VITAMIN A which functions as the active component of the visual cycle. It is the prosthetic group of RHODOPSIN (i.e., covalently bonded to ROD OPSIN as 11-cis-retinal). When stimulated by visible light, rhodopsin transforms this cis-isomer of retinal to the trans-isomer (11-trans-retinal). This transformation straightens-out the bend of the retinal molecule and causes a change in the shape of rhodopsin triggering the visual process. A series of energy-requiring enzyme-catalyzed reactions convert the 11-trans-retinal back to the cis-isomer.
