The regulated release of proinsulin from secretory granules accompanied by cleavage of proinsulin to form mature insulin. In vertebrates, insulin is secreted from B granules in the B cells of the vertebrate pancreas and from insulin-producing cells in insects. [GOC:mah, ISBN:0198506732]
Insulin secretion is a complex process that is tightly regulated by blood glucose levels. It involves a series of events that begin with the entry of glucose into pancreatic beta cells. Glucose is transported into the cells via the GLUT2 transporter. Once inside the cell, glucose is metabolized through glycolysis and the citric acid cycle, leading to the production of ATP. The increased ATP levels close ATP-sensitive potassium (KATP) channels in the plasma membrane of the beta cells. This closure of KATP channels depolarizes the membrane potential, opening voltage-gated calcium (Ca2+) channels. The influx of Ca2+ triggers the fusion of insulin-containing secretory granules with the plasma membrane, releasing insulin into the bloodstream. This process is known as exocytosis. Insulin secretion is also regulated by other factors, including glucagon-like peptide-1 (GLP-1), glucose-dependent insulinotropic polypeptide (GIP), and incretins. These factors enhance insulin secretion by increasing cAMP levels and activating protein kinase A (PKA). PKA phosphorylates and activates proteins involved in insulin secretion, leading to a greater release of insulin. In summary, insulin secretion is a complex and tightly regulated process that is initiated by the entry of glucose into pancreatic beta cells. This triggers a series of events, including the closure of KATP channels, depolarization of the membrane potential, opening of Ca2+ channels, and exocytosis of insulin-containing granules. Insulin secretion is also regulated by other factors, such as incretins, which amplify the insulin response to glucose.'
