excitatory synapse

Definition

Target type: cellularcomponent

A synapse in which an action potential in the presynaptic cell increases the probability of an action potential occurring in the postsynaptic cell. [GOC:dph, GOC:ef]

The excitatory synapse is a specialized junction between neurons where one neuron, the presynaptic neuron, releases neurotransmitters that stimulate the postsynaptic neuron. The cellular component of an excitatory synapse is comprised of several distinct structures:

**Presynaptic Terminal:**

* **Synaptic Vesicles:** These small, membrane-bound organelles contain neurotransmitters, such as glutamate, which are the signaling molecules of excitatory synapses.
* **Active Zones:** Specialized regions of the presynaptic membrane where synaptic vesicles dock and release their neurotransmitters.
* **Presynaptic Density:** A dense protein scaffold that anchors synaptic vesicles, active zones, and other signaling molecules.

**Synaptic Cleft:**

* **Extracellular Matrix:** A complex mixture of proteins and carbohydrates that fills the space between the pre- and postsynaptic neurons and helps to maintain the structural integrity of the synapse.
* **Neurotransmitter Receptors:** These proteins are embedded in the postsynaptic membrane and bind to neurotransmitters released from the presynaptic terminal, triggering a signal in the postsynaptic neuron.

**Postsynaptic Density:**

* **Postsynaptic Receptors:** These proteins are concentrated in the postsynaptic membrane and bind to neurotransmitters released from the presynaptic terminal. They include ionotropic receptors, which allow ions to flow across the membrane, and metabotropic receptors, which activate intracellular signaling pathways.
* **Signaling Molecules:** A variety of proteins that interact with postsynaptic receptors and regulate the strength and duration of synaptic transmission.
* **Scaffolding Proteins:** These proteins provide structural support for the postsynaptic density and help to organize the various signaling molecules.

**Other Components:**

* **Mitochondria:** Provide energy for synaptic transmission.
* **Microtubules and Actin Filaments:** These cytoskeletal elements help to transport vesicles and maintain the structural integrity of the synapse.

The precise composition and organization of the cellular components of an excitatory synapse can vary depending on the type of synapse, the neurons involved, and the function of the synapse. However, the basic principles of synaptic transmission, involving the release of neurotransmitters from the presynaptic terminal and their binding to receptors on the postsynaptic membrane, are conserved across different types of excitatory synapses.'
