neutrophil chemotaxis

Definition

Target type: biologicalprocess

The directed movement of a neutrophil cell, the most numerous polymorphonuclear leukocyte found in the blood, in response to an external stimulus, usually an infection or wounding. [GOC:jl, ISBN:0198506732]

Neutrophil chemotaxis is a crucial process in the innate immune response, involving the directed migration of neutrophils towards sites of infection or tissue damage. This process is initiated by the release of chemoattractants, such as chemokines, bacterial products, and complement components, from the injured tissue or invading pathogens.

These chemoattractants bind to specific receptors on the surface of neutrophils, triggering a complex signaling cascade within the cell. This cascade leads to the activation of various intracellular pathways, including the PI3K-Akt and MAPK pathways, which ultimately result in the polymerization of actin filaments. Actin polymerization is essential for the formation of pseudopodia, finger-like projections that extend from the cell's leading edge.

Neutrophils move directionally towards the source of chemoattractants by a process called chemotaxis. This involves the coordinated extension and retraction of pseudopodia, with the cell migrating in the direction of the highest concentration of chemoattractant.

The movement of neutrophils is guided by a series of biochemical events, including the activation of G-protein-coupled receptors, the production of second messengers like cAMP and calcium ions, and the activation of various kinases and phosphatases.

As neutrophils migrate towards the site of injury, they encounter various adhesion molecules on the surface of endothelial cells, such as ICAM-1 and VCAM-1. These interactions allow neutrophils to adhere to the vessel wall and eventually transmigrate through the endothelium into the surrounding tissue.

Once neutrophils have reached the site of infection, they release various antimicrobial substances, such as reactive oxygen species, proteases, and antimicrobial peptides. These substances contribute to the killing of pathogens and the clearance of debris from the injured tissue.

In summary, neutrophil chemotaxis is a multi-step process that involves the sensing of chemoattractants, the activation of intracellular signaling pathways, the formation of pseudopodia, the directed migration towards the chemoattractant source, and the release of antimicrobial substances at the site of infection.'
