lipoprotein transport

Definition

Target type: biologicalprocess

The directed movement of any conjugated, water-soluble protein in which the nonprotein group consists of a lipid or lipids, into, out of or within a cell, or between cells, by means of some agent such as a transporter or pore. [GOC:jl, ISBN:0198506732]

Lipoprotein transport is a complex process that involves the movement of lipids, including cholesterol, triglycerides, and phospholipids, throughout the body. It is essential for delivering these nutrients to cells for energy production, membrane synthesis, and hormone production. Here is a detailed description of the process:

1. **Lipid Absorption and Packaging:** Dietary lipids, primarily triglycerides and cholesterol, are absorbed in the small intestine. Within the intestinal cells, these lipids are packaged into chylomicrons, which are large, triglyceride-rich lipoprotein particles.

2. **Chylomicron Transport:** Chylomicrons enter the lymphatic system and eventually reach the bloodstream. As they circulate, lipoprotein lipase (LPL), an enzyme found on the surface of capillary endothelial cells, hydrolyzes triglycerides in the chylomicrons, releasing free fatty acids for uptake by tissues.

3. **Chylomicron Remnant Formation:** After triglyceride hydrolysis, the chylomicron remnants, enriched in cholesterol, are taken up by the liver.

4. **Hepatic Lipoprotein Synthesis:** The liver synthesizes various lipoprotein particles, including very-low-density lipoproteins (VLDL), low-density lipoproteins (LDL), and high-density lipoproteins (HDL).

5. **VLDL Transport:** VLDL particles transport newly synthesized triglycerides and cholesterol from the liver to peripheral tissues. Similar to chylomicrons, LPL hydrolyzes VLDL triglycerides, releasing free fatty acids and resulting in VLDL remnants.

6. **LDL Formation:** VLDL remnants are further metabolized to LDL, which is the primary carrier of cholesterol in the bloodstream. LDL delivers cholesterol to cells for membrane synthesis and hormone production.

7. **HDL Formation:** HDL is synthesized in the liver and intestine. It functions as a "scavenger" of cholesterol, picking up excess cholesterol from cells and tissues and transporting it back to the liver for excretion or re-utilization.

8. **Reverse Cholesterol Transport:** The process of HDL transporting cholesterol from peripheral tissues to the liver is known as reverse cholesterol transport.

The balance and efficiency of lipoprotein transport are critical for maintaining healthy lipid levels and preventing cardiovascular disease. Dysregulation of lipoprotein metabolism can lead to high LDL levels, which contribute to atherosclerosis, while low HDL levels are associated with increased risk of heart disease.'

Thyroxine is synthesized via the iodination of tyrosines (MONOIODOTYROSINE) and the coupling of iodotyrosines (DIIODOTYROSINE) in the THYROGLOBULIN. Thyroxine is released from thyroglobulin by proteolysis and secreted into the blood. Thyroxine is peripherally deiodinated to form TRIIODOTHYRONINE which exerts a broad spectrum of stimulatory effects on cell metabolism.

Although some is produced in the thyroid, most of the 3,3',5-triiodo-L-thyronine in the body is generated by mono-deiodination of L-thyroxine in the peripheral tissues. Its metabolic activity is about 3 to 5 times that of L-thyroxine. The sodium salt is used in the treatment of hypothyroidism.

Triiodothyronine: A T3 thyroid hormone normally synthesized and secreted by the thyroid gland in much smaller quantities than thyroxine (T4). It is a major active constituent of the seed oil of Croton tiglium. It has been used as a tumour promoting agent for skin carcinogenesis in rodents and is associated with increased cell proliferation of malignant cells. However its function is controversial since a decrease in cell proliferation has also been observed in several cancer cell types.

The compound is a potent inhibitor of HIV replication, acting as a chain-terminator of viral DNA during reverse transcription. It improves immunologic function, partially reverses the HIV-induced neurological dysfunction, and improves certain other clinical abnormalities associated with AIDS. mycophenolic acid : A member of the class of 2-benzofurans that is 2-benzofuran-1(3H)-one which is substituted at positions 4, 5, 6, and 7 by methyl, methoxy, (2E)-5-carboxy-3-methylpent-2-en-1-yl, and hydroxy groups, respectively. It is an antibiotic produced by Penicillium brevi-compactum, P. stoloniferum, P. echinulatum and related species. An immunosuppressant, it is widely used (partiularly as its sodium salt and as the 2-(morpholin-4-yl)ethyl ester prodrug, mycophenolate mofetil) to prevent tissue rejection following organ transplants and for the treatment of certain autoimmune diseases.

Mycophenolic Acid: Compound derived from Penicillium stoloniferum and related species. It blocks de novo biosynthesis of purine nucleotides by inhibition of the enzyme inosine monophosphate dehydrogenase (IMP DEHYDROGENASE). Mycophenolic acid exerts selective effects on the immune system in which it prevents the proliferation of T-CELLS, LYMPHOCYTES, and the formation of antibodies from B-CELLS. It may also inhibit recruitment of LEUKOCYTES to sites of INFLAMMATION.

It is a prodrug for the corresponding sulfide, a non-steroidal anti-inflammatory drug, used particularly in the treatment of acute and chronic inflammatory conditions.

Sulindac: A sulfinylindene derivative prodrug whose sulfinyl moiety is converted in vivo to an active NSAID analgesic. Specifically, the prodrug is converted by liver enzymes to a sulfide which is excreted in the bile and then reabsorbed from the intestine. This helps to maintain constant blood levels with reduced gastrointestinal side effects.
