The generation of amyloid-beta by cleavage of the amyloid precursor protein (APP). [GOC:mah]

Amyloid-beta (Aβ) formation is a complex biological process that involves the sequential processing of the amyloid precursor protein (APP) by two key enzymes: β-secretase (BACE1) and γ-secretase. APP is a transmembrane protein that is normally found on the surface of neurons and other cells in the brain.

The process begins with the cleavage of APP by BACE1, which is a transmembrane aspartic protease. This cleavage occurs within the extracellular domain of APP, generating a soluble N-terminal fragment (sAPPα) and a membrane-bound C-terminal fragment (C99).

Next, C99 is processed by γ-secretase, a multi-protein complex that includes presenilin, nicastrin, APH-1, and PEN-2. γ-secretase is a complex enzyme that can cleave C99 at multiple sites, generating different Aβ peptides. The most common forms of Aβ are Aβ40 and Aβ42, which differ in length by two amino acids. Aβ42 is more prone to aggregation than Aβ40 and is considered to be the most toxic form of Aβ.

Aβ peptides are normally cleared from the brain by various mechanisms, including degradation by enzymes and removal from the brain by the lymphatic system. However, when these clearance mechanisms are impaired, Aβ can accumulate in the brain, forming amyloid plaques. These plaques are believed to be a major contributor to the development of Alzheimer's disease.

The precise mechanism by which Aβ plaques contribute to Alzheimer's disease is not fully understood. However, it is thought that the accumulation of Aβ in the brain can trigger a cascade of events that lead to neuronal dysfunction and death. These events may include inflammation, oxidative stress, and the formation of neurofibrillary tangles, which are another hallmark of Alzheimer's disease.
