The series of molecular signals initiated by glutamate binding to an NMDA-selective glutamate receptor on the surface of the target cell, followed by the movement of ions through a channel in the receptor complex, and ending with the regulation of a downstream cellular process, e.g. transcription. The NMDA receptor (NMDAR) is a glutamate receptor that plays a crucial role in synaptic plasticity, learning, and memory. It is a ligand-gated ion channel that is permeable to sodium, potassium, and calcium ions. The NMDAR is unique in that it requires the binding of both glutamate and glycine to open the channel. In addition, it is voltage-dependent, meaning that it requires a certain level of membrane depolarization to become fully active. When glutamate binds to the NMDAR, the channel opens, allowing calcium ions to flow into the postsynaptic neuron. Calcium is a second messenger that activates a variety of intracellular signaling pathways, including the activation of protein kinases, such as CaMKII and PKC. These kinases can phosphorylate and activate other proteins, leading to changes in gene expression, synaptic strength, and neuronal morphology. The NMDAR is also involved in the regulation of neuronal survival and apoptosis. Overactivation of the NMDAR can lead to excitotoxicity, a process that can damage and kill neurons. This process is thought to be involved in neurodegenerative diseases such as Alzheimer's disease, Parkinson's disease, and Huntington's disease. The NMDAR is a complex and highly regulated protein. Its activity is modulated by a variety of factors, including the presence of other neurotransmitters, the concentration of intracellular calcium, and the phosphorylation state of the receptor. The NMDAR is a critical component of the nervous system and plays a vital role in a variety of brain functions. Its dysfunction is associated with a number of neurological and psychiatric disorders.'
