The modification of an organic chemical by the conjugation of glucuronic acid. The substances resulting from glucuronidation are known as glucuronosides (or glucuronides) and are often much more water-soluble than the non-glucuronic acid-containing precursor. [GOC:BHF]
Glucuronidation is a critical detoxification and elimination process in the liver and other tissues, involving the conjugation of glucuronic acid, a sugar molecule, to various substrates, including steroids, bilirubin, drugs, and hormones. This process occurs within the endoplasmic reticulum, catalyzed by a family of enzymes called UDP-glucuronosyltransferases (UGTs).
The process begins with the activation of glucuronic acid, derived from glucose, by UDP-glucuronic acid (UDPGA). This activated form is then transferred to the substrate by UGTs, forming a glucuronide conjugate. The substrate molecule is typically hydrophobic, rendering it difficult to eliminate from the body. However, glucuronidation transforms the substrate into a more hydrophilic molecule, enhancing its water solubility and facilitating its excretion through the kidneys or bile.
