The MyD88-dependent toll-like receptor (TLR) signaling pathway is a crucial component of the innate immune system, playing a vital role in recognizing and responding to invading pathogens. This pathway is activated by a diverse range of microbial components, including bacterial lipopolysaccharide (LPS), viral double-stranded RNA (dsRNA), and fungal zymosan.

The initiation of the MyD88-dependent TLR signaling pathway begins with the binding of a specific microbial ligand to its cognate TLR on the cell surface or in endosomes. TLRs are transmembrane receptors that possess an extracellular domain responsible for ligand recognition and an intracellular Toll/IL-1 receptor (TIR) domain. The TAK1 complex, composed of TAK1, TAB1, and TAB2, is a crucial signaling module that activates downstream pathways. TAK1 is a serine/threonine kinase that phosphorylates both IκB kinase (IKK) and MAPK kinases (MKKs).

The activation of IKK leads to the phosphorylation and subsequent degradation of IκBα, an inhibitor of the transcription factor nuclear factor kappa B (NF-κB). The release of NF-κB allows its translocation to the nucleus, where it triggers the transcription of a wide range of pro-inflammatory genes, including cytokines, chemokines, and adhesion molecules.

Simultaneously, the activation of MKKs by TAK1 results in the phosphorylation of MAP kinases, including ERK, JNK, and p38. These MAP kinases are involved in various cellular processes, including inflammation, cell survival, and apoptosis.
