Endocannabinoid-mediated trans-synaptic signaling is a crucial mechanism regulating neuronal activity, influencing synaptic plasticity and contributing to various physiological functions. This process involves the release of endocannabinoids, lipid messengers synthesized on demand, from postsynaptic neurons in response to neuronal activity. The primary endocannabinoids, anandamide (AEA) and 2-arachidonoylglycerol (2-AG), are produced by different enzymes, with AEA synthesized by N-acylphosphatidylethanolamine phospholipase D (NAPE-PLD) and 2-AG by diacylglycerol lipase (DAGL). Once released, endocannabinoids diffuse across the synaptic cleft and bind to cannabinoid receptors (CB1) predominantly located on presynaptic terminals. CB1 activation triggers a signaling cascade involving the Gi/o protein and the inhibition of adenylyl cyclase, leading to a decrease in intracellular cAMP levels. This, in turn, reduces the activity of protein kinase A (PKA), ultimately leading to a dampening of presynaptic neurotransmitter release. The modulation of synaptic transmission by endocannabinoids is highly context-dependent, acting as a negative feedback mechanism to control neuronal activity. This feedback regulation can influence various aspects of synaptic plasticity, including long-term potentiation (LTP) and long-term depression (LTD).
