The chemical reactions and pathways involving norepinephrine, a hormone secreted by the adrenal medulla, and a neurotransmitter in the sympathetic peripheral nervous system and in some tracts in the central nervous system. It is also the demethylated biosynthetic precursor of epinephrine. [GOC:jl, ISBN:0198506732]
Norepinephrine, a neurotransmitter and hormone, undergoes a complex metabolic process to regulate its levels in the body. The primary enzyme responsible for norepinephrine breakdown is monoamine oxidase (MAO), which converts norepinephrine to normetanephrine. This reaction occurs in the cytoplasm of neurons and other cells, and it involves the removal of an amine group from the norepinephrine molecule. MAO exists in two forms, MAO-A and MAO-B, with MAO-A being the primary enzyme involved in norepinephrine metabolism.
Another pathway for norepinephrine breakdown involves catechol-O-methyltransferase (COMT), which converts norepinephrine to metanephrine. This reaction occurs in the cytosol of neurons and other cells and involves the addition of a methyl group to the norepinephrine molecule.
In addition to these primary metabolic pathways, norepinephrine can also be re-uptaken into presynaptic neurons by the norepinephrine transporter (NET). Once inside the presynaptic neuron, norepinephrine can be repackaged into synaptic vesicles for release or it can be metabolized by MAO or COMT.
The regulation of norepinephrine metabolism is crucial for maintaining normal neurotransmission and physiological function. Dysregulation of norepinephrine metabolism can lead to a variety of disorders, including depression, anxiety, and attention-deficit/hyperactivity disorder (ADHD). Medications that inhibit MAO or COMT activity can be used to increase norepinephrine levels in the brain, which can be beneficial for treating these disorders.'
