A protein complex containing one Bcl protein and one or more copies of NF-kappaB2; formation of complexes of different stoichiometry depends on the Bcl3:NF-kappaB2 ratio, and allow Bcl3 to exert different regulatory effects on NF-kappaB2-dependent transcription. [GOC:mah, PMID:9407099]
The Bcl3/NF-kappaB2 complex is a transcription factor complex that plays a role in the regulation of gene expression. The cellular component of this complex consists of the following components:
1. **Bcl3:** Bcl3 is a member of the IkappaB family of proteins, which are known to inhibit the activity of NF-kappaB. However, Bcl3 acts as a co-activator of NF-kappaB, rather than an inhibitor. Bcl3 binds to the p50/p65 heterodimer of NF-kappaB, and this interaction is essential for the transcriptional activity of the complex.
2. **NF-kappaB2 (p52/p100):** NF-kappaB2 is a heterodimer composed of the p52 and p100 subunits. p100 is a precursor protein that is processed by the proteasome to generate the active p52 subunit. p52 can form a heterodimer with RelA (p65), RelB, or c-Rel, but it is typically associated with Bcl3.
3. **DNA:** The Bcl3/NF-kappaB2 complex binds to specific DNA sequences, known as kappaB sites, in the promoters of target genes. These kappaB sites are typically located within 50 bp of the transcription start site.
4. **Other proteins:** In addition to Bcl3, NF-kappaB2, and DNA, the Bcl3/NF-kappaB2 complex may also interact with other proteins, such as histone acetyltransferases (HATs), which are involved in the regulation of chromatin structure.
