Any process that modulates the rate, frequency, or extent of the mitotic cell cycle spindle assembly checkpoint, a cell cycle checkpoint that delays the metaphase/anaphase transition of a mitotic nuclear division until the spindle is correctly assembled and chromosomes are attached to the spindle. The spindle assembly checkpoint (SAC) is a critical regulatory mechanism that ensures accurate chromosome segregation during mitosis. This checkpoint monitors the proper attachment of microtubules from the spindle poles to kinetochores, protein complexes assembled on centromeres of chromosomes.
The SAC functions through a surveillance system that involves a complex interplay of proteins, including:
**1. Sensor Proteins:** These proteins, such as Mad1, Mad2, and Bub1, are localized at kinetochores and monitor the status of microtubule attachment.
**2. Signaling Proteins:** When a kinetochore is unattached or improperly attached, sensor proteins activate signaling proteins like Mad2 and BubR1. These proteins form a complex that inhibits the anaphase promoting complex/cyclosome (APC/C).
**3. APC/C Inhibition:** The APC/C is a ubiquitin ligase that targets key regulatory proteins for degradation, including securin and cyclin B. Inactivation of APC/C by the SAC prevents the activation of separase, an enzyme that cleaves cohesin, the protein responsible for holding sister chromatids together.
**4. Checkpoint Maintenance:** As long as unattached or improperly attached kinetochores remain, the SAC remains active, preventing the onset of anaphase and cell cycle progression.
**5. Checkpoint Release:** Once all kinetochores are correctly attached to microtubules, the signal is relayed back to the sensor proteins, leading to a decrease in the inhibition of APC/C. This allows for the activation of separase, leading to the cleavage of cohesin, and the separation of sister chromatids.
