photoreceptor disc membrane

Definition

Target type: cellularcomponent

Stack of disc membranes located inside a photoreceptor outer segment, and containing densely packed molecules of photoreceptor proteins that traverse the lipid bilayer. Disc membranes arise as evaginations of the ciliary membrane during the development of the outer segment and may or may not remain contiguous with the ciliary membrane. [GOC:bj, GOC:krc, GOC:pde, PMID:11826267, PMID:19501669, PMID:2537204, PMID:26574505, PMID:6771304, PMID:7507907]

The photoreceptor disc membrane is a specialized membrane structure found in the outer segments of photoreceptor cells, the light-sensitive cells in the retina. It is responsible for capturing light and converting it into electrical signals that are transmitted to the brain.

The disc membrane is composed of a phospholipid bilayer that is enriched in specific lipids and proteins. The lipids provide the structural framework of the membrane, while the proteins carry out the key functions of light absorption and signal transduction.

**Key components of the photoreceptor disc membrane:**

* **Rhodopsin:** A light-sensitive G protein-coupled receptor (GPCR) that is embedded in the disc membrane. Rhodopsin absorbs light and triggers a cascade of signaling events that lead to the generation of an electrical signal.
* **Transducin:** A G protein that is activated by rhodopsin. Activated transducin activates phosphodiesterase, an enzyme that breaks down cyclic GMP (cGMP).
* **Phosphodiesterase:** An enzyme that hydrolyzes cGMP, leading to a decrease in cGMP levels in the photoreceptor cell. This decrease in cGMP levels closes sodium channels in the plasma membrane, leading to a hyperpolarization of the photoreceptor cell.
* **Cyclic GMP (cGMP):** A second messenger molecule that is involved in the regulation of sodium channels in the plasma membrane of photoreceptor cells.
* **Other proteins:** The disc membrane also contains other proteins, such as arrestin, which binds to rhodopsin and deactivates it, and recoverin, which regulates the activity of transducin.

**Organization of the disc membrane:**

The disc membrane is organized into stacks of flattened discs that are separated by a narrow interdisc space. The discs are formed by the invagination of the plasma membrane of the outer segment. The discs are continuously renewed, with new discs being formed at the base of the outer segment and older discs being shed at the tip.

**Function of the disc membrane:**

The photoreceptor disc membrane plays a crucial role in vision by capturing light and converting it into electrical signals. This process begins when light strikes rhodopsin, causing a conformational change in the protein. This conformational change activates transducin, which in turn activates phosphodiesterase. Phosphodiesterase breaks down cGMP, leading to a decrease in cGMP levels and a hyperpolarization of the photoreceptor cell. This hyperpolarization signal is transmitted to the brain, where it is interpreted as a visual signal.'
