ATP generation from poly-ADP-D-ribose

The process of generating ATP in the nucleus from poly-ADP-D-ribose. Nuclear ATP generation is required for extensive chromatin remodeling events that are energy-consuming. [PMID:27257257]

Poly-ADP-ribose (PAR) is a branched polymer of ADP-ribose units, synthesized by poly(ADP-ribose) polymerases (PARPs) from NAD+. PARylation is a dynamic post-translational modification that plays a crucial role in various cellular processes, including DNA repair, gene transcription, and apoptosis. The breakdown of PAR, catalyzed by poly(ADP-ribose) glycohydrolase (PARG), is crucial for maintaining cellular homeostasis. While PAR is not directly involved in ATP generation, its breakdown through PARG can indirectly contribute to ATP levels. PARG utilizes water to cleave the glycosidic bond between ADP-ribose units in PAR, releasing free ADP-ribose. This ADP-ribose can then be recycled through salvage pathways to generate NAD+, which is the precursor for ATP synthesis. This process, however, is not a significant source of ATP compared to oxidative phosphorylation, the main pathway for ATP production. Therefore, while PARG activity can indirectly influence ATP levels through NAD+ recycling, the primary function of PAR degradation is not ATP generation, but rather to maintain cellular homeostasis and regulate PARylation-mediated cellular processes.'
