Target type: molecularfunction
Catalysis of the reaction: L-tyrosyl-[protein] + NAD+ = H+ + nicotinamide + O-(ADP-D-ribosyl)-L-tyrosyl-[protein]. [PMID:29954836, PMID:30257210, RHEA:58236]
NAD+-protein-tyrosine ADP-ribosyltransferase activity is a molecular function associated with enzymes that catalyze the transfer of an ADP-ribose moiety from NAD+ to a tyrosine residue on a target protein. This process, known as ADP-ribosylation, involves the formation of a covalent bond between the ADP-ribose group and the tyrosine side chain. NAD+-protein-tyrosine ADP-ribosyltransferases play crucial roles in cellular signaling, protein regulation, and various physiological processes. The enzymatic activity of these proteins is highly specific, targeting certain tyrosine residues within specific protein substrates. The ADP-ribosylation of these tyrosine residues can alter the conformation, activity, or interactions of the target protein. This modification can serve as a post-translational regulatory mechanism, modulating the protein's function and contributing to diverse cellular responses. NAD+-protein-tyrosine ADP-ribosyltransferase activity is involved in a range of cellular processes, including DNA repair, cell cycle regulation, and immune responses. Dysregulation of this enzymatic activity has been linked to various diseases, including cancer and neurodegenerative disorders.'
