Substance P receptor activity is a molecular function associated with the G protein-coupled receptor (GPCR) known as the neurokinin-1 receptor (NK1R). This receptor is activated by the neuropeptide substance P, which plays a significant role in various physiological processes, including pain perception, inflammation, neurotransmission, and mood regulation. The binding of substance P to NK1R triggers a cascade of intracellular signaling events. The receptor undergoes conformational changes, leading to the activation of G proteins, primarily Gq, which in turn activates phospholipase C (PLC). PLC hydrolyzes phosphatidylinositol 4,5-bisphosphate (PIP2) into diacylglycerol (DAG) and inositol trisphosphate (IP3). DAG activates protein kinase C (PKC), while IP3 triggers the release of calcium from intracellular stores.
