11-beta-hydroxysteroid dehydrogenase (NADP+) activity catalyzes the interconversion of cortisol and cortisone. This enzymatic activity plays a critical role in regulating the intracellular concentration of active glucocorticoids.

Specifically, the enzyme reduces cortisone to cortisol, the active form of the hormone, in tissues that express the enzyme, such as the kidney, liver, and adipose tissue. Conversely, it oxidizes cortisol to cortisone, the inactive form, in other tissues, such as the brain.

This dynamic balance between cortisol and cortisone is crucial for maintaining normal glucocorticoid signaling and regulating various physiological processes, including metabolism, inflammation, and immune responses.
