Fatty acid synthase activity refers to the enzymatic process of synthesizing fatty acids de novo from acetyl-CoA and malonyl-CoA as building blocks. This multi-step process occurs within a large, multi-domain enzyme complex called fatty acid synthase (FAS). FAS catalyzes a series of reactions that ultimately result in the elongation of a growing fatty acid chain. The process begins with the attachment of acetyl-CoA to the acyl carrier protein (ACP) domain of FAS. Malonyl-CoA is then added to the growing chain, and a series of reductions, dehydrations, and reductions are performed. This cycle of reactions is repeated until the desired chain length is reached. The final product is a saturated fatty acid, typically palmitate (C16:0). Fatty acid synthase activity is essential for the biosynthesis of essential fatty acids, which are necessary for membrane formation, energy storage, and signaling processes. This activity is also critical for the production of various lipids, such as triglycerides, phospholipids, and cholesterol.
