calcitonin gene-related peptide receptor signaling pathway

Definition

Target type: biologicalprocess

The series of molecular signals initiated by an extracellular calcitonin gene-related peptide (CGRP) combining with a calcitonin gene-related peptide receptor on the surface of the target cell. Calcitonin gene-related peptide receptors may form dimers, trimers or tetramers. [GOC:bhm, PMID:10882736]

The calcitonin gene-related peptide (CGRP) receptor signaling pathway is a complex cascade of events triggered by the binding of CGRP to its receptor. CGRP is a neuropeptide involved in various physiological processes, including vasodilation, pain perception, and inflammation. The CGRP receptor is a heterodimer composed of two subunits: calcitonin receptor-like receptor (CLR) and receptor activity-modifying protein 1 (RAMP1).

When CGRP binds to the CLR/RAMP1 receptor, it activates a G protein called Gαs. This activation leads to the production of cyclic adenosine monophosphate (cAMP) through the activation of adenylyl cyclase. Increased cAMP levels activate protein kinase A (PKA), which phosphorylates various downstream targets.

One of the key targets of PKA is the transcription factor CREB (cAMP response element-binding protein). Phosphorylated CREB binds to the cAMP response element (CRE) in the promoter regions of genes, leading to their transcriptional activation.

The CGRP receptor signaling pathway also interacts with other signaling pathways, such as the mitogen-activated protein kinase (MAPK) pathway. Activation of the MAPK pathway leads to the phosphorylation of MAPKs, which further contribute to gene expression and cell proliferation.

In addition to activating downstream signaling pathways, CGRP receptor signaling can also lead to the release of other signaling molecules, such as nitric oxide (NO) and prostaglandins. NO is a potent vasodilator, while prostaglandins play a role in inflammation and pain perception.

Overall, the CGRP receptor signaling pathway is a complex and highly regulated process that plays a crucial role in various physiological functions. Understanding this pathway has led to the development of new therapeutic targets for treating conditions such as migraine and chronic pain.'
