Tie signaling pathway

The series of molecular signals initiated by an angiopoietin binding to the Tie receptor, and ending with the regulation of a downstream cellular process, e.g. transcription. [GOC:ceb, GOC:signaling, PMID:11283723, PMID:11566266]

The Tie signaling pathway is a crucial regulator of vascular development and maintenance. It involves two receptor tyrosine kinases, Tie1 and Tie2, and their respective ligands, angiopoietin-1 (Ang1) and angiopoietin-2 (Ang2).

Tie2, the primary receptor in this pathway, plays a central role in vascular stability and maturation. It is expressed primarily on endothelial cells, the cells lining blood vessels. Ang1, the primary ligand for Tie2, promotes the formation of stable and mature blood vessels. Upon Ang1 binding to Tie2, the receptor undergoes dimerization and autophosphorylation, activating downstream signaling cascades. These cascades involve various signaling molecules, including phospholipase C gamma (PLC-γ), phosphoinositide 3-kinase (PI3K), and mitogen-activated protein kinases (MAPKs). These signaling events lead to the activation of transcription factors, such as the nuclear factor kappa B (NF-κB), which control gene expression involved in vascular development and maintenance. Ang1 signaling through Tie2 promotes endothelial cell survival, proliferation, and migration, contributing to vessel stabilization and integrity.

Ang2, the second ligand for Tie2, acts as a competitive antagonist of Ang1. In the absence of Ang1, Ang2 binds to Tie2 and induces its internalization and degradation, leading to a decrease in Tie2 signaling. This downregulation of Tie2 signaling allows for vascular remodeling and sprouting, processes that are necessary for angiogenesis, the formation of new blood vessels. During angiogenesis, Ang2 promotes endothelial cell detachment and sprouting by counteracting the stabilizing effects of Ang1.

Tie1, the second receptor in the Tie signaling pathway, is also expressed on endothelial cells. While its exact function remains under investigation, it is believed to act as a modulator of Tie2 signaling. It can bind to both Ang1 and Ang2 but with lower affinity compared to Tie2. Tie1 may cooperate with Tie2 in promoting vascular stability or act as a negative regulator of Tie2 signaling under certain conditions.
