DNA-dependent protein kinase (DNA-PK) activity plays a crucial role in DNA double-strand break (DSB) repair, specifically the non-homologous end joining (NHEJ) pathway. This pathway is essential for maintaining genome integrity and preventing mutations. DNA-PK is a serine/threonine protein kinase that is activated by the presence of DNA DSBs. The activation process involves the binding of DNA-PK catalytic subunit (DNA-PKcs) to DNA ends, followed by autophosphorylation and recruitment of other repair factors. Once activated, DNA-PK performs several key functions in NHEJ: 1) It recruits and phosphorylates the Ku70/Ku80 heterodimer, which binds to DNA ends and protects them from degradation. 2) It facilitates the recruitment and activation of other repair factors, such as the Artemis nuclease, which processes DNA ends for ligation. 3) It promotes the alignment of broken DNA ends, facilitating their ligation by the DNA ligase IV complex. 4) It interacts with other DNA repair pathways, including homologous recombination, to ensure efficient DSB repair.
