A protein complex that consists of three clathrin heavy chains and three clathrin light chains, organized into a symmetrical three-legged structure called a triskelion. In clathrin-coated vesicles clathrin is the main component of the coat and forms a polymeric mechanical scaffold on the vesicle surface. [GOC:mah, PMID:16493411]

The clathrin complex is a protein complex that plays a crucial role in endocytosis, the process by which cells internalize molecules and particles from their environment. It is a large, cage-like structure composed of three major protein components: clathrin, adaptor proteins (APs), and accessory proteins.

Clathrin, a triskelion-shaped protein, forms the structural framework of the complex. It is composed of three heavy chains and three light chains that assemble into a lattice-like structure. Each triskelion consists of three legs, each with a terminal domain that interacts with adaptor proteins.

Adaptor proteins (APs) mediate the interaction between clathrin and cargo. They bind to specific sorting signals on cargo molecules and to clathrin, linking the cargo to the clathrin coat. There are four major types of AP complexes: AP-1, AP-2, AP-3, and AP-4, each responsible for sorting different types of cargo.

Accessory proteins are a diverse group of proteins that regulate the assembly and disassembly of the clathrin coat. They include proteins that promote the formation of clathrin-coated pits, proteins that stabilize the coat, and proteins that promote the uncoating of the vesicle.

The clathrin complex assembles at specific sites on the plasma membrane, forming clathrin-coated pits. Cargo molecules that are destined for internalization are concentrated in these pits. As the clathrin coat assembles, the membrane invaginates, forming a vesicle. The vesicle then detaches from the plasma membrane and is transported to the appropriate intracellular compartment.

The clathrin complex plays a critical role in many cellular processes, including:

* Removal of cellular debris and pathogens
* Regulation of cell signaling
* Trafficking of proteins within the cell
