Target type: molecularfunction
Catalysis of the reaction: NADP+ + 5,6,7,8-tetrahydropteridine = NADPH + H+ + 6,7-dihydropteridine. [EC:1.5.1.34]
6,7-dihydropteridine reductase activity catalyzes the NADPH-dependent reduction of 7,8-dihydrobiopterin to tetrahydrobiopterin. This enzyme plays a crucial role in the biosynthesis of tetrahydrobiopterin, a cofactor essential for several enzymatic reactions, including the hydroxylation of phenylalanine to tyrosine by phenylalanine hydroxylase, the hydroxylation of tyrosine to dopa by tyrosine hydroxylase, and the oxidation of tryptophan to 5-hydroxytryptophan by tryptophan hydroxylase. These reactions are involved in the synthesis of neurotransmitters such as dopamine, norepinephrine, epinephrine, and serotonin. 6,7-dihydropteridine reductase activity also participates in the regeneration of tetrahydrobiopterin from dihydrobiopterin, which is formed during the catalytic cycle of the aforementioned monooxygenases. Mutations in the gene encoding this enzyme can lead to various disorders, including hyperphenylalaninemia, dopa-responsive dystonia, and neurotransmitter deficiencies.'
