A protein complex that is involved in the repair of DNA double-strand breaks and, in mammals, V(D)J recombination events. It consists of the DNA-dependent protein kinase catalytic subunit (DNA-PKcs) and the DNA end-binding heterodimer Ku. [GOC:mah, PMID:10854421, PMID:12235392]
The DNA-dependent protein kinase (DNA-PK) complex is a multi-protein complex involved in DNA double-strand break (DSB) repair by non-homologous end joining (NHEJ). It is composed of three subunits:
- **DNA-PKcs:** The catalytic subunit of the complex. It is a large, 460 kDa protein with a phosphoinositide 3-kinase (PI3K)-like domain and a DNA-binding domain. It possesses intrinsic DNA-dependent protein kinase activity, which is required for the phosphorylation of target proteins during DSB repair.
- **Ku70/Ku80 heterodimer:** This heterodimer binds to the ends of DSBs and acts as a scaffold for the recruitment of DNA-PKcs to the break site. It provides DNA-PKcs with access to the DNA ends and protects them from degradation.
- **XRCC4/Ligase IV complex:** This complex is responsible for the ligation of the broken DNA ends after they have been processed and aligned by the DNA-PK complex.
The DNA-PK complex plays a crucial role in NHEJ repair. It performs the following functions:
- **Binding to DNA ends:** The Ku70/Ku80 heterodimer recognizes and binds to the DNA ends, providing a platform for the recruitment of DNA-PKcs.
- **Activation of DNA-PKcs:** Once bound to the DNA ends, DNA-PKcs undergoes conformational changes and becomes activated, enabling it to phosphorylate target proteins.
- **Phosphorylation of target proteins:** Activated DNA-PKcs phosphorylates various proteins involved in DSB repair, including itself, Ku70/Ku80, and other repair factors. This phosphorylation event helps to regulate the repair process.
- **Recruitment of other repair factors:** The phosphorylated DNA-PKcs complex acts as a signaling hub, recruiting additional repair factors to the DSB site.
- **Processing of DNA ends:** The DNA-PK complex facilitates the processing of the broken DNA ends, preparing them for ligation.
- **Ligating the broken DNA ends:** The XRCC4/Ligase IV complex, which is recruited to the DSB site by DNA-PK, performs the final step of joining the processed DNA ends.
