Target type: molecularfunction
(S)-dihydroorotate + a quinone = orotate + a quinol. [RHEA:30187]
Dihydroorotate dehydrogenase (quinone) activity catalyzes the oxidation of dihydroorotate to orotate, a crucial step in the de novo biosynthesis of pyrimidine nucleotides. This enzyme utilizes a quinone as an electron acceptor, which is then reduced to a hydroquinone. The reaction occurs in the mitochondrial matrix and involves the transfer of two electrons from dihydroorotate to the quinone. The oxidized form of the quinone, such as ubiquinone (CoQ), acts as a mobile electron carrier in the mitochondrial electron transport chain, contributing to ATP synthesis. Dihydroorotate dehydrogenase plays a pivotal role in the synthesis of pyrimidine nucleotides, which are essential building blocks for DNA and RNA. Mutations in the gene encoding this enzyme can lead to orotic aciduria, a rare genetic disorder characterized by an accumulation of orotic acid in the urine and severe anemia. The enzyme's activity is regulated by the availability of its substrates, dihydroorotate and quinone, as well as by the levels of its products, orotate and reduced quinone.'
