2-aminoadipate transaminase activity

Definition

Target type: molecularfunction

Catalysis of the reaction: 2-oxoglutarate + L-2-aminoadipate = 2-oxoadipate + L-glutamate. [EC:2.6.1.39, RHEA:12601]

2-aminoadipate transaminase activity catalyzes the transfer of an amino group from an amino donor, such as glutamate, to 2-aminoadipate, resulting in the formation of 2-oxoadipate and the corresponding amino acid. This reaction is essential in the lysine degradation pathway, where it converts 2-aminoadipate, an intermediate in lysine catabolism, to 2-oxoadipate, which can then be further metabolized. The reaction proceeds through a ping-pong mechanism, where the enzyme first binds the amino donor and releases it after transferring the amino group to a pyridoxal phosphate cofactor. The resulting pyridoxamine phosphate form then binds 2-aminoadipate and transfers the amino group to it, generating 2-oxoadipate and regenerating the pyridoxal phosphate form of the enzyme. This catalytic cycle ensures efficient conversion of 2-aminoadipate to 2-oxoadipate, contributing to the overall degradation of lysine. 2-aminoadipate transaminase activity is also involved in the biosynthesis of certain amino acids, such as pipecolate, which is a precursor for the production of other essential compounds.'
