Target type: cellularcomponent
The component of a membrane consisting of gene products and protein complexes that are loosely bound to one of its surfaces, but not integrated into the hydrophobic region. [GOC:dos, GOC:jl, GOC:mah]
The extrinsic component of the membrane, also known as the peripheral membrane protein, is a type of protein that associates with the cell membrane without actually embedding itself within the phospholipid bilayer. Unlike integral membrane proteins that span the entire membrane, extrinsic proteins interact with the membrane surface via a variety of mechanisms:
* **Electrostatic interactions:** These proteins possess charged amino acid residues that interact with the polar head groups of phospholipids, forming ionic bonds.
* **Hydrophobic interactions:** Some extrinsic proteins contain hydrophobic patches that interact with the nonpolar fatty acyl chains of membrane lipids.
* **Interactions with integral membrane proteins:** They can bind to the exposed domains of integral membrane proteins, either on the cytoplasmic or extracellular side of the membrane.
Extrinsic proteins are typically located on the outer leaflet of the plasma membrane, interacting with the extracellular environment, or on the inner leaflet, associated with the cytoskeleton. They play a crucial role in various cellular functions:
* **Signal transduction:** They act as receptors for extracellular signals, transmitting information into the cell.
* **Cell adhesion:** They mediate cell-cell and cell-matrix interactions, contributing to tissue formation and organization.
* **Enzyme activity:** Some extrinsic proteins exhibit enzymatic activity, participating in metabolic processes.
* **Cytoskeletal anchoring:** They link the plasma membrane to the underlying cytoskeleton, maintaining cell shape and providing structural integrity.
Their non-covalent association with the membrane allows them to be readily dissociated by changes in pH, salt concentration, or the presence of specific agents. This dynamic nature allows for their regulated recruitment and removal from the membrane, contributing to their diverse cellular roles.'
