ATP-dependent protein disaggregases are molecular chaperones that play a critical role in maintaining cellular proteostasis by resolving protein aggregates. These aggregates can form due to various factors, including stress conditions, mutations, and aging, and can disrupt cellular function. ATP-dependent protein disaggregases utilize the energy from ATP hydrolysis to unfold and disassemble protein aggregates, thereby restoring the functional state of the proteins. The process involves several steps: First, the disaggregase binds to the aggregate and uses its ATPase activity to generate mechanical force. This force helps to pull apart the aggregated proteins, separating them into individual polypeptide chains. Next, the disaggregase can either refold the unfolded proteins into their native conformations or deliver them to other chaperones for further processing. Some disaggregases, like the Hsp104 family, can even assist in the disaggregation of amyloid fibrils, which are highly stable and often associated with neurodegenerative diseases. The molecular mechanism of disaggregation involves the coordinated action of several domains within the disaggregase molecule. The ATPase domain provides energy for the process, while other domains, like the substrate-binding domain, interact with the aggregated proteins.
