Deoxyhypusine monooxygenase (DHPS) catalyzes the conversion of deoxyhypusine to hypusine, a unique amino acid found in eukaryotic translation initiation factor 5A (eIF5A). Hypusine is essential for the function of eIF5A, which is involved in a variety of cellular processes, including translation elongation, cell proliferation, and stress response. The mechanism of the reaction catalyzed by DHPS involves the oxidation of the deoxyhypusine residue by molecular oxygen to form hypusine. This oxidation is a two-step process that involves the formation of a reactive intermediate, which is then reduced by NADPH.
