2-C-methyl-D-erythritol 2,4-cyclodiphosphate synthase (MCS) catalyzes the formation of 2-C-methyl-D-erythritol 2,4-cyclodiphosphate (MEcPP) from 4-diphosphocytidyl-2-C-methyl-D-erythritol (CDP-ME). This reaction is a key step in the non-mevalonate pathway, which is an alternative pathway for the biosynthesis of isopentenyl pyrophosphate (IPP) and dimethylallyl pyrophosphate (DMAPP), essential building blocks for the synthesis of terpenes, steroids, and other isoprenoids. The enzyme utilizes a two-step mechanism. First, MCS binds CDP-ME and Mg2+ ions, which are required for catalysis. Then, MCS catalyzes the intramolecular cyclization of CDP-ME, resulting in the formation of a cyclic phosphate ring. This is followed by the release of CMP (cytidine monophosphate), leading to the formation of MEcPP. This enzyme plays a critical role in the biosynthesis of isoprenoids, which are essential molecules in various biological processes, including photosynthesis, respiration, and hormone synthesis.
