6-phosphofructo-2-kinase (PFK-2) activity is a key regulatory step in glycolysis and gluconeogenesis. It catalyzes the phosphorylation of fructose 6-phosphate to fructose 2,6-bisphosphate (F2,6BP). This reaction is crucial for the regulation of carbohydrate metabolism as F2,6BP acts as a potent allosteric activator of phosphofructokinase 1 (PFK-1), the rate-limiting enzyme of glycolysis, and an inhibitor of fructose 1,6-bisphosphatase (FBPase-1), the rate-limiting enzyme of gluconeogenesis. In essence, PFK-2 activity promotes glycolysis by enhancing PFK-1 activity and suppressing FBPase-1 activity, leading to increased glucose utilization and ATP production.
