AnxA2-p11 complex

Definition

Target type: cellularcomponent

A heterotetrameric protein complex comprising two Annexin A2 (AnxA2) monomers and two copies of its binding partner, S100 protein p11 (S100A10). [GOC:bf, GOC:BHF, GOC:nc, PMID:18799458, PMID:23483454]

The AnxA2-p11 complex is a protein complex that plays a critical role in the regulation of intracellular calcium signaling and membrane trafficking. The cellular component of this complex is primarily localized to the plasma membrane, where it interacts with various signaling molecules and membrane-associated proteins.

AnxA2, also known as annexin A2, is a calcium-dependent phospholipid-binding protein that exhibits a characteristic "butterfly" structure with four conserved domains. Its interaction with p11, a protein also known as S100A10, is crucial for the formation and stability of the AnxA2-p11 complex.

The localization of the AnxA2-p11 complex to the plasma membrane is mediated by the binding of p11 to the N-terminal domain of AnxA2. This interaction promotes the recruitment of AnxA2 to the membrane, where it can interact with other proteins involved in signaling pathways, such as phospholipase A2 (PLA2) and the tyrosine kinase Src.

The AnxA2-p11 complex also associates with specific membrane domains, including lipid rafts, which are specialized microdomains enriched in cholesterol and sphingolipids. This association with lipid rafts allows the complex to interact with signaling molecules and membrane-associated proteins that are localized within these microdomains.

In addition to its localization at the plasma membrane, the AnxA2-p11 complex has also been reported to be present in other cellular compartments, such as the Golgi apparatus and endosomes. However, its primary function and localization are associated with the plasma membrane, where it plays a critical role in regulating cellular processes related to calcium signaling, membrane trafficking, and cell adhesion.'
