[Hydroxyurea--is it a harmless drug in Vaquez disease?]].

    The frequent side effects of Hydroxy-Urea and the non-exceptional risk of leukemia and cancer in Polycythemia Vera treated for a long time by Hydroxy-Urea allow to conclude that Hydroxy-Urea is not an innocent drug. In a prospective trial of 150 patients with a median follow up of nine years, Hydroxy-Urea given alone induced side effects in 29% of patients necessitating to stop treatment in half of cases. The percentage of leukemia or myelodysplasia is 6.7% with an actuarial risk of leukemic transformation of 10% at 13 years. In an other prospective trial in 181 aged patients Hydroxy-Urea was given as maintenance therapy after 32P treatment. The median follow-up in that study is also of nine years. Side effects are observed in 13% of cases. A two fold increase of the leukemic risk was observed in the maintenance arm of the trial: 11 versus 19% at ten years, 14 vs 30% at 12 years, 16 vs 35% at 15 years because of the leukemogenic effect of Hydroxy-Urea in maintenance therapy we stopped including new patients in this arm of the trial.

[Tumor progression as a factor of variability of diurnal rhythm of phosphorus metabolism in a tumor].

    In a transplanted lymphosarcoma of rats by means of Cosinor method we discovered rhythm of P32 inclusions with ultradian and circadian periods. In the course of tumor growth changes of spectrum of periods, amplitudes and acrophases of rhythms take place. These shifts are explained by the progression and clonal structure of neoplasms.

Hormonal regulation of transcription of rDNA: use of nucleoside thiotriphosphates to measure initiation in isolated nuclei.

    Nuclei isolated from cultured mouse and rat cell lines initiated pre-rRNA chains at the correct site and with the correct nucleotide specificity (A for mouse, G for rat). Nucleic acid filter hybridization and S1 nuclease mapping were used to analyze the RNA products initiated with nucleoside (beta-S)triphosphates. Initiation of pre-rRNA was completely resistant to alpha-amanitin but was inhibited by either actinomycin D or heparin. Experiments with P1798 mouse lymphoma cells indicated that the antiproliferative effects of glucocorticoids on lymphoid cells includes a reduction in the ability of nuclei to initiate pre-rRNA chains.

[Circadian rhythm of adenosine triphosphatase activity and 32P content in Pliss lymphosarcoma].

    Within a daily period maximal activity of ATPases and content of 32P in Pliss lymphosarcoma tissue was found in the samples taken at 15 or 21 p.m. and 3 a.m. The radioactivity in the tumor was minimal between 9 and 12 a.m. and 18 or 24 p.m. The alterations in Mg2+-ATPase activity in their magnitude and circadian rhythm coincided completely with the daily variations in content of radiophosphorus. Activities of Na+, K+- and HCO(3-)-ATPases exhibited maximal values at 15 p.m. and, especially between 3 and 6 a.m., i.e. these alterations were of two-peak shape without the peak at 21 p.m.
