The constitutional isomers and tautomers of oxadiazolones, as well as their mono- and disulfur analogues, were calculated at the B3LYP/aug-cc-pVDZ level. Four groups of 30 molecules each were considered: oxadiazolone, oxadiazolthione, thiadiazolone, and thiadiazolthione isomers. The compounds were categorized into six groups according to permutations of three heteroatoms in the five-membered ring. Additionally, each of the constitutional isomer was considered to have five tautomers conserving stable five-membered ring: two NH tautomers, two rotameric OH (or SH) forms and one CH. Novel lipid-polymer hybrid nanoparticles incorporated in thermosensitive in situ gel for intranasal delivery of terbutaline sulphate.
The present work aimed to improve the bioavailability of terbutaline sulphate (TS) and to prolong its nasal residence time for the treatment of asthma.. Chitosan/pectin polyelectrolyte complex nanoparticles (CS/PC) were prepared by ionic gelation method and coated with phospholipid (PL) and then incorporated into optimised thermosensitive in situ gel.. The optimal PL-coated nanoparticle formulation (LP1) showed the smallest particle size (345.5 nm), the highest zeta potential (32.9 mV) and the greatest percent drug released after 6 h (71%). The optimum in situ gel loaded with LP1 (NG3) showed three times greater permeation through nasal mucosa than aqueous solution of TS and revealed about 94% and 92% of the effect of IV injection of drug solution on tidal volume and peak expiratory flow in histamine treated rats, respectively.. The developed PL-coated CS/PC/in situ gel could be considered as a promising intranasal formulation of TS for asthma management. Microbiota Contribute to Obesity-related Increases in the Pulmonary Response to Ozone.
Obesity is a risk factor for asthma, especially nonatopic asthma, and attenuates the efficacy of standard asthma therapeutics. Obesity also augments pulmonary responses to ozone, a nonatopic asthma trigger. The purpose of this study was to determine whether obesity-related alterations in gut microbiota contribute to these augmented responses to ozone. Dietary supplementation with specific oligosaccharide mixtures decreases parameters of allergic asthma in mice.
Specific mixtures of prebiotic oligosaccharides showed immune modulatory effects in previous murine vaccination experiments, suggesting a shift towards T-helper 1 (Th1) immunity. These mixtures consisted of short-chain galacto-oligosaccharides (scGOS) and long-chain fructo-oligosaccharides (lcFOS) in a 9:1 ratio (Immunofortis), with or without pectin-derived acidic oligosaccharides (pAOS). To investigate whether these mixtures could suppress Th2-related responses, they were tested in an ovalbumin (OVA)-induced model for experimental allergic asthma in BALB/c mice. Supplementation with two mixtures of scGOS/lcFOS and scGOS/lcFOS/pAOS at approximately 1% (w/w% net oligosaccharides) in the diet, starting two weeks before OVA sensitization and lasting until the end of the experiment, decreased of several parameters of allergic asthma. The OVA-induced airway inflammation and hyperresponsiveness was significantly suppressed by both mixtures. Moreover, OVA-specific IgE titers were decreased by more than 25%, although this effect was not significant. The effects of the oligosaccharide mixture with pAOS appeared to be more pronounced than the effects of the scGOS/lcFOS mixture without pAOS, but a direct comparison between the mixtures was not made. Overall, the results further support the hypothesis that specific mixtures of oligosaccharides modulate the Th1/Th2 balance by enhancing Th1-related and suppressing Th2-related parameters. Asian pear pectin administration during presensitization inhibits allergic response to ovalbumin in BALB/c mice.
A type of respiratory disorder resembling some aspects of human allergic asthma can be induced in mice using ovalbumin. The factors that influence the etiology of asthma are poorly understood even though cytokines are known to play a pivotal role. The purpose of this study was to test the hypothesis whether an administration of Asian pear pectin during presensitization could suppress allergic response to ovalbumin in BALB/c mice.. High-dose (100 microg) of pectin-sol was used and values were compared to those from the control. Ovalbumin and aluminum hydroxide were utilized for sensitization while ovalbumin aerosol was used for provocation 2 weeks later. The bronchoalveolar lavage (BAL) and assessment of tracheal smooth muscle responsiveness to electrical field stimulation or acetylcholine were performed 1 day after ovalbumin provocation. Two main cytokines of interferon (IFN)-gamma and interleukin (IL)-5, and serum immunoglobulin E (IgE) were assayed.. Laboratory of the Chosun University Medical School. Male BALB/c mice. Antigen dose of 5 microg for sensitization generated TH1 type cytokines in the lungs with a high level of IFN-gamma and a low level of IL-5. In contrast, TH2 type cytokines were produced in splenocytes including a high level of IL-5 and a low level of IFN-gamma. Asian pear pectin-sol administration during presensitization significantly inhibited (p < 0.05) sensitivity of airway smooth muscle to electrical field stimulation and acetylcholine. Further, IFN-gamma production significantly decreased (p < 0.05) in BAL fluids while it significantly increased (p < 0.05) in splenic cells. On the other hand, IL-5 production significantly increased (p < 0.05) in BAL fluids while it was a significant decrease (p < 0.05) in splenic cells. For the histopathologic changes in the lung, pear pectin-sol recovered ovalbumin (OVA)-induced abnormal signs to an almost normal state. As a correlate, IgE production significantly decreased (p < 0.05) in pectin-sol-treated animals compared to the control.. It is possible from these data that BALB/c mice have different susceptibilities to different doses of OVA regulated by pulmonary TH1 and TH2 type cytokines, independent of splenic TH1 and TH2 type cytokines production. These results also indicate that administration of Asian pear pectin-sol in presensitized mice suppresses allergic asthmatic reaction. Occupational asthma caused by pectin inhalation during the manufacture of jam.
We report a case of pectin-induced occupational asthma in a 35-year-old man. His job involved mixing powdered pectin into a fruit puree during the manufacture of jam. Within minutes of adding pectin, he developed coryza, rhinorrhea, coughing, and wheezing. His symptoms cleared during weekends while away from work and improved with the use of a protective face mask at work. Peak flow rates were significantly lower while at work compared with those at home, and a prick skin test with the pectin powder was positive. We conclude that pectin should be added to the list of the substances known to induce occupational asthma. Christmas candy maker's asthma. IgG4-mediated pectin allergy.
We evaluated a 29-year-old candy maker with no history of asthma who developed asthma after exposure to pectin, a compound manufactured from fruits and fruit rinds. Following eight years of employment during which he added pectin to a recipe for Christmas candies, the candymaker developed acute respiratory symptoms. Challenge testing with the pectin mixture caused a 40 percent decrease in FEV1. Skin prick testing was positive to the pectin extract. Total IgE was normal and pectin-specific IgE antibodies were not detected. A strongly positive pectin-specific IgG4 antibody response was present that was not detected in a control serum and could be inhibited by the addition of pectin. Antigen-specific IgG4 should be sought in IgE negative cases of occupational asthma.
