Comparison of the effector functions of human immunoglobulins using a matched set of chimeric antibodies.
Cell lines have been established that secrete a matched set of human chimeric IgM, IgG1, IgG2, IgG3, IgG4, IgE, and IgA2 antibodies that are directed against the hapten 4-hydroxy-3-nitrophenacetyl. These chimeric antibodies secreted from mouse plasmacytoma cells behave exactly like their authentic human counterparts in SDS-PAGE analysis, binding to protein A and in a wide range of serological assays. The antibodies have been compared in their ability to bind human C1q as well as in their efficacy in mediating lysis of human erythrocytes in the presence of human complement. A major conclusion to emerge is that whereas IgG3 bound C1q better than did IgG1, the chimeric IgG1 was much more effective than all the other IgG subclasses in complement-dependent hemolysis. The IgG1 antibody was also the most effective in mediating antibody-dependent cell-mediated cytotoxicity using both human effector and human target cells. These results suggest that IgG1 might be the favoured IgG subclass for therapeutic applications. Stimulation of cells by antibody.
Tumor cell lines exposed to immunoglobulins specific for cell surface antigens developed increased cellular incorporation of [(125)I]iododeoxyuridine and [(3)H]thymidine (up to 200-fold increases over cells treated with normal rabbit immunoglobulins). Antibody-stimulated cells multiplied more rapidly and lived longer than control cells in tissue culture. These observations were made both with cells substituted with 2,4,6-trinitrophenol and purified antibody against 2,4,6-trinitrophenol, and with several cell lines and their respective whole-cell antibodies. Antibodies that were stimulatory at low concentrations were cytotoxic at high concentrations. These observations may have significance in regard to enhancing effects of antibodies on tumor cell growth in vivo. Autoimmune-like antibodies to the ligand-binding sites of myeloma proteins.
BALB/c mice were immunized with three A-myeloma proteins of BALB/c-2 or BALB/c origin (produced by plasmacytomas MOPC-315, MOPC-460, Adj. PC-22A). Noncross reacting antibodies were formed against Proteins-315 and 460, but the response to Protein-22A was marginal. Proteins-315 and 460 have anti-dinitrophenyl activity, and their reactions with the corresponding BALB/c antibodies were inhibited by dinitrophenyl ligands. It appears that antibodies can be formed in BALB/c mice against unique "idiotypic" determinants in the ligand-binding sites of some BALB/c mycloma proteins.
