Menaquinone (vitamin K2) therapy for bronchial asthma. II. Clinical effect of menaquinone on bronchial asthma.

    A series of 191 patients with bronchial asthma were treated with menaquinone for not less than one year to investigate the clinical effects of the drug. The clinical effect may be summarized as follows: 1) The therapy with menaquinone only gave an effective rate oo 90.9% (a markedly effective rate of 42.4%) in mild patients, an effective rate of 86.7% (a markedly effective rate of 30.0%) in moderate patients, and effective rate of 72.7% (a markedly effective rate of 27.3%) in severe patients; namely, considerably a high effective rate was attained by the therapy in the respective patients. 2) The therapy with menaquinone, in conjunction with hyposensitization therapy, gave an effective rate of 100% (a markedly effective rate of 33.3%). 3) The double blind study of the clinical effects of menaquinone revealed that placebo used in the study was effective on only 16.7% of the patients treated with it, and that the incidence of recurrence due to withdrawal of meaquinone therapy tended to be low in the patients treated with the drug for a long period of time.

The relationship between childhood leukaemia and childhood asthma: A pharmacoepidemiological study from the Netherlands.

    It has been suggested that childhood asthma lowers the risk of childhood leukaemia. Studies have found an inverse association between these conditions. However, most studies on this relationship are based on questionnaires and telephone interviews, introducing recall bias. Therefore, we conducted a matched case-control study based on drug prescription data to assess the relationship between both conditions.. In a large database, covering more than one million individuals, we identified cases of children who had been prescribed 6-mercaptopurine (6-MP). This drug is used in the outpatient maintenance therapy of childhood leukaemia. We matched every child with leukaemia on sex and age (±6 months) to children without leukaemia (controls). The variable of having had asthma was defined as receiving at least two prescriptions for an inhaled corticosteroid within 12 months.. We identified 59 children aged 2-18 who had been prescribed 6-MP (cases), and they were matched to 21,918 controls. Of the children with childhood leukaemia, three (5%) had childhood asthma, whereas in the control group 4889 (22%) had childhood asthma (odds ratio [OR] 0.19; 95% confidence interval 0.06-0.60).. In this study on the relationship between childhood asthma and childhood leukaemia, we found a strong inverse association.

Clinically relevant pharmacogenomic testing in pediatric practice.

The objective of personalized medicine is to improve healthcare by tailoring disease prevention, diagnosis, and treatment strategies for individuals based on their unique clinical history and genetic composition. This article offers an overview of pharmacogenomics, discusses caveats specific to pharmacogenomics in pediatric populations, provides evidence-based recommendations for pediatric clinicians, and offers insight regarding the future role of pharmacogenomics testing in pediatric medicine. Reviews of the current literature and thoughtful discussions are presented regarding the pharmacogenomics of antidepressants, codeine and oncologic, asthma, and immunomodulatory pharmacotherapies.
