[Studies on the anticancer effect of howiinol A, a new compound isolated from Goniothalamus howii].
The effect of howiinol A (GHM-10) on the growth of several cancer and normal cells were studied, using the methods of cell growth curve determination, MTT test and soft-agar colony formation assay. The results showed that GHM-10 exhibited potent inhibitory effect on cancer cells with an IC50 of 2 micrograms.ml-1 approximately. Normal cells, especially bone marrow progenitor cells, were shown to be less sensitive to GHM-10. A drug-resistant cell line, KB/VCR200, was found to be as sensitive as its parental KB cells. GHM-10 did not induce HL-60 cell differentiation. In murine transplantable tumors, GHM-10 was found to have significant therapeutic effect on mice bearing solid tumor hepatoma H22, Lewis lung carcinoma and S180 ascites. [The chemical constituents of Goniothalamus howii Merr].
Two new compounds were isolated from the chloroform soluble fraction of ethanolic extract of bark of Goniothalamus howii Merr. On the basis of their chemical properties and spectral data (MS, UV, IR, 1H and 13CNMR) they were identified as 6S (1R-hydroxy-2R-cinnamyloxyphenethyl) 5, 6-dihydro-5S-hydroxy-2-pyrone named Howiinol A and 6S-(1S, 2R-epoxyphenethyl-5S-cinnamyloxy) 5, 6-dihydro-2-pyrone named Howiinin A. Howiinol A showed significant antitumor activities toward human tumor cell in vitro and in vivo and less toxic.
