Ginsenoside Rg3 promotes regression from hepatic fibrosis through reducing inflammation-mediated autophagy signaling pathway.
Inflammation and autophagy occur during hepatic fibrosis development caused by various pathogens, and effectively curbing of autophage may delay the occurrence of hepatic fibrosis. The current study aimed to unravel the inhibitory effects of Ginsenoside Rg3 (G-Rg3) on inflammation-mediated hepatic autophagy to curb hepatic fibrosis caused by thioacetamide (TAA)-induced subacute and chronic hepatic injury. TAA is mainly metabolized in the liver to cause liver dysfunction. After intraperitoneal injection of TAA for 4 or 10 weeks (TAA-chronic mouse models), severe inflammatory infiltration and fibrosis occurred in the liver. Treatment with G-Rg3 alleviated hepatic pathological changes and reversed hepatic fibrosis in the TAA-chronic models with decreased deposition of collagen fibers, reduced expression of HSCs activation marker (α-SMA), and reduced secretion of profibrogenic factors (TGF-β1). G-Rg3 decreased expressions of autophagy-related proteins in mice of TAA-chronic models. Notably, G-Rg3 inhibited the survival of activated rat hepatic stellate cells (HSC-T6), but had no cytotoxicity on human hepatocytes (L02 cell lines). G-Rg3 dose-dependently inhibited autophagy in vitro with less expression of p62 and fewer LC3a transformation into LC3b in inflammatory inducer lipopolysaccharide (LPS)-induced rat HSC-T6 cells. Furthermore, G-Rg3 enhanced the phosphorylation of phosphatidylinositol 3-kinase (PI3K) and protein kinase B (Akt) in vivo and in vitro. Besides, mTOR inhibitor Rapamycin and PI3K inhibitors LY294002 were employed in LPS-treated HSC-T6 cell cultures to verify that Rg3 partially reversed the increase in autophagy in hepatic fibrosis in vitro. Taken together, G-Rg3 exerted anti-fibrosis effect through the inhibition of autophagy in TAA-treated mice and LPS-stimulated HSC-T6 cells. These data collectively unravel that G-Rg3 may serve a promising anti-hepatic fibrosis drug. Pharmaceutical Efficacy of Gypenoside LXXV on Non-Alcoholic Steatohepatitis (NASH).
Ginsenosides have offered a wide array of beneficial roles in the pharmacological regulation of hepatic metabolic syndromes, including non-alcoholic steatohepatitis (NASH), non-alcoholic fatty liver disease (NAFLD), and obesity. Of the numerous ginsenosides, Rg3 has been widely investigated, but there have been few studies of gypenosides (Gyp). Particularly, no study on Gyp LXXV has been reported to date. Here, to firstly explore the pharmacological effects of Gyp LXXV against NASH and the related mechanism, methionine- and choline-deficient (MCD) diet-induced NASH mice and hepatic cells (stellate cells, hepatic macrophages, and hepatocytes) were selected. Gyp LXXV exhibited markedly alleviated MCD diet-induced hepatic injury, inflammation, and fibrosis by down-regulating hepatic fibrosis markers such as α-smooth muscle actin(α-SMA), collagen1, transforming growth factors-β (TGF-β1), tumor necrosis factor-α (TNF-α), MCP-1, interleukin (IL)-1β, nuclear factor κB (NFκB), and GRP78. Remarkably, histopathological studies confirmed that 15 mg/kg of Gyp LXXV administration to MCD diet-induced mice led to effective prevention of liver injury, lipid accumulation, and activation of hepatic macrophages, indicating that Gyp LXXV might be a potential anti-NASH drug. [Effect of ginsenoside Rg3 on hepatic fibrosis in murine schistosomiasis japonica].
To investigate the therapeutic efficacy of ginsenoside Rg3 on hepatic fibrosis in murine schistosomiasis japonica.. 54 ICR-strain male mice were divided into 4 groups named as normal control group (A), infected control group (B), praziquantel+Rg3 treated group (C) and praziquantel treated group (D). There were 12 mice in each group, but 18 in group A. Mice in groups B, C, and D were infected with 20 +/- 2 cercariae of Schistosoma japonicum. At ten weeks post-infection, 10 mice of group A and 12 mice of group B were weighed and sacrificed. Specimens from left hepatic lobes were taken and fixed in 10% formaldehyde solution. Mice in groups C and D were treated intragastrically with praziquantel at a single dose of 300 mg/kg. At the second day after praziquantel treatment, each mouse in group C was given 3 mg/(kg x d) ginsenoside Rg3 for 8 weeks. The rest mice were sacrificed on 8 weeks after treatment, and liver tissue samples from left hepatic lobes were prepared. The histological changes and collagen fiber deposition in the liver tissue sections were observed with hematoxylin-eosin staining and van gieson staining. Liver fibrosis was graded according to semi-quantitative scoring system (SSS) method.. In group B, many eggs deposited in the hepatic lobules and portal areas, and eosinophilic abscesses and pseudo-tubercles developed in the liver, especially common in portal areas. There were many fibre hyperplasia and deposit inside abbacy and liver flocculus. Pipestem fibrosis formed around the portal areas, and some cord-like fibres extended into hepatic lobules, and formed in the fibrous septa. After 8-week treatment with ginsenoside Rg3, in group C, the livers were initially enlarged, firm and dust-color; and the degree of hepatomegaly varied from mild to marked; but the degree of fibre hyperplasia and inflammatory infiltration were mitigated compared with that of group B. Mean percentage of collagen area in group C [(2.32 +/- 0.99)%] was lower than that of groups B [(11.08 +/- 4.43)%] and D [(11.19 +/- 4.91)%] (P < 0.05). The SSS scores of hepatic fibrosis in group C (2.83 +/- 1.09) was lower than that of groups B (7.42 +/- 1.16) and D (8.08 +/- 1.76) (P < 0.05)..
