[Soil dust as a factor which changes the histochemical picture of the rat lung].

    White Wistar female rats only once were administered intratracheally a suspension of the soil dust in the physiologic saline. The respirable fraction of the soil dust was used for preparing the suspension of the latter. The experiment was carried out in 2 groups of rats. The animals of the 1st group received the soil dust suspension which was the mixture of the soil samples from 3 different places in Poland, the animals of the 2nd group from 3 other places in Poland. The control animals were once administered the physiologic saline. 9 months after the application of dust, the specimen of the left lung were taken for histochemical examinations. The reactions to the activity of succinic and lactate dehydrogenase, of tetrazole reductase NADP, and of acid phosphatase and ATP-ase were performed. The changes in the activity of the studied respiratory enzymes and the increase of the reaction to the acid phosphatase were observed. The results reveal the disorders of the oxidation-reduction processes and the intensification of the lytic processes in the pulmonary parenchyma of the animals exposed to the action of the soil dust.

[Histochemical mechanisms in etiology of pulmonary fibrosis after short-term exposure to a mixture of dust and quartz from copper mine].

    The authors induced pulmonary silicosis in albino rats by the intratracheal method with 50 mg mixed middling slime from Panaguiriste and quartz dust, dissolved in I ml physiological solution. The animals received a single treatment and were killed on 90th day of the experiment. Some intime characteristics of collagen-forming mechanisms in lungs were followed up via routine histological, histochemical, enzymohistochemical/ acid phosphatase activity, beta-glucoronidase, AS-esterase and lipase/ and autoradiographic/H3 labelled proline/ methods. It was established, that under the effect of both dusts from Panaguiriste mines studied/ mixed dust and pure quartz dust/ considerable deviations developed in the structure of the lungs of the experimental animals in the enzyme activity of acid phosphatase and carbon esterase, accompanied by various forms of fibrosis in the organ, depending on the dust applied. The difference in the composition of both dusts quartz and mixed dust slime - had an effect on the degree of the deviations, but not on their character, manifested in: I. specific protective reaction of organism, with activation of the ferments from the group of carbon esterase in macrophagic elements of interstitium and around the silicotic alterations in the lungs of the experimental animals; 2. inhibition of the maturation of macrophages in silicotic foci and 3. acceleration of the proliferation of fibroblasts with intensification of collagen-formation, accompanied by an enhanced activity of the ferments from the group of carbon esterase and in the cytoplasm of fibroblasts. The authors draw the conclusion that the complex use of routine histological, histochemical and quantitative isotope methods enables the more significant determination of fibrinogenicity of quartz-containing dust even at the early terms of their exposure under experimental conditions.

[Structuro-functional coniophage transformation as a response to fibrogenic dust and the possibility of its medicinal correction].

    Electron microscopic and cytochemical studies indicate that structural and functional changes in coniophage occur under the influence of fibrogenous dust. These changes consiste in fragmentation of single phagolysosomes containing dust particles with the release of the contents into the cytoplasm, disorders in the structure of mitochondria, granular endoplasmic reticulum, and plate apparatus. These changes lead to a decline in the bioenergetics and biosynthetic function of coniophage. At a late stage, a damage of phagolysosomal apparatus as a whole causes autolysis of the cell. A principal possibility of directed effect on the membrane structures of coniophage for the purpose of influencing the initial mechanisms of silicosis development is demonstrated.
