[Anti-cancer activity of Zuojinwan in vivo and influence to tumor markers in mice transplanted with sarcoma 180].

    To study the anticancer action of Zuojinwan in mice transplanted with sarcoma 180 in vivo, and detect the activities of five kinds of tumor markers (TM) including acid phosphotase (ACP), alkaline phosphotase (AKP), creatine kinase (CK), aldolase (ALD) and lactate dehydrogenase (LDH) in serum compared with Coptis chinensis and Evodia rutaecarpa.. The transplanted S180 tumor mice model was established, and the mice were divided randomly five groups. The extract of Zuojinwan (850.8 mg kg(-1)), C. chinensis (729.2 mg kg(-1)) and E. rutaecarpa (121.6 mg kg(-1)) were administrated, respectively for 10 d. Then, the changes of body weight, spleen index of mice, the inhibition rates of tumor, and the increase of life span (ILS) were all tested. In addition, the activities of ACP, AKP, CK, ALD and LDH on different test groups were also determined.. Zuojinwan could inhibit the S180 tumor growth significantly with the inhibition rate of 50.54% and the ILS of mice reached to 64.91%. Meanwhile, the activities of ACP (126.72 +/- 11.16) U 100 mL(-1) and AKP (67.27 +/- 13.49) U 100 mL(-1) were increased, and the activities of CK (20.65 +/- 4.28) U mL(-1), ALD (319.13 +/- 53.87) U L(-1) and LDH (1,029.04 +/- 468.56) U L(-1) were decreased significantly by Zuojinwan treated group compared with C. chinensis and E. rutaecarpa treated groups (P<0.01).. In the prescription of Zuojinwan, the enhancement of compatibility of anticancer activity was observed by the interaction of C. chinensis and E. rutaecarpa. The mechanism might be in according with to influence the activities of the five kinds of tumor markers (TM) in mice serum.

Ascites sarcoma 180, an animal model of humoral hypercalcemia of malignancy, produces a factor(s) exhibiting potent bone-resorbing activity without any parathyroid hormone-like activity.

    Ascites sarcoma 180 (S180A) is a transplantable tumor maintained in ddY mice. In the tumor-bearing mice, the plasma Ca, Pi and acid phosphatase levels increased and the plasma alkaline phosphatase levels decreased. The elevation of plasma Pi levels is unusual in humoral hypercalcemia of malignancy (HHM). To characterize the pathogenesis of HHM in the animals, the biological activities in the serum-free conditioned media (CM) of S180A cell cultures were examined. The S180A CM stimulated bone resorption dose dependently and showed TGF-like, IL-1-like and mitogenic activity. Unlike parathyroid hormone (PTH), the factor(s) failed to stimulate cAMP production by either UMR 106-01 cells or neonatal mouse calvaria at concentrations that stimulate bone resorption. Also, the factor(s) stimulated proliferation of UMR 106-01 cells concomitant with a slight increase in intracellular calcium levels. These results indicate that S180A cells produce a factor(s) responsible for bone resorption which is apparently different from PTH-like activity.

Enzyme content of hepatocellular lysosomes in the liver of the tumour-bearing mouse.

    An increase in the number of hepatocellular lysosomes was not detected in mice bearing the solid or ascites forms of Sarcoma 180 or Ehrlich's tumour. Despite this, biochemical studies showed significant increases in acid phosphatase, cathepsin-D and beta-galactosidase. This difference between morphological and biochemical observations is explained by the fact that only gross increases in lysosome population are likely to be detected in the small samples examined with the electron microscope, but by biochemical methods whereby many grams of tissue are analysed finer changes may be detected. As far as is known at present there is an increase in hepatocellular lysosomes and/or lysosomal enzymes in tumour-bearing mice, rats and man and no exception to this exists.

Enzymatic responses of transplanted tumour cells towards estrogen, progesterone and testosterone.

    The influence of estrogen, progesterone and testosterone on the activities of alkaline and acid phosphatases, adenosine triphosphatase and succinate dehydrogenase were determined by cytochemical methods in sarcoma 180 and Ehrlich's carcinoma cells transplanted in male and female Swiss mice. The results revealed differential effects of the sex hormones on different enzymes which seemed to depend on the type of tumour cell studied and the sex of the host mice.. Cells of sarcoma 180 and of Ehrlich's carcinoma were maintained by serial transplantation in male and female Swiss mice. Either estrogen, progesterone, or testosterone were injected im at doses of 1 mg/mouse. Ascitic fluid was aspirated at intervals of 1, 3, 6, 24, and 48 hours following hormone injections. Enzyme activities were analyzed by subjective grading according to the intensity of staining reaction. Estrogen produced enhancement of alkaline phosphatase activity in both types of cells in both sexes of mice. Progesterone produced increased alkaline phosphatase activity in both types of cells from female hosts but an inhibitory effect in male hosts' cells. Testosterone produced no change in enzyme activity in tumor cells of female hosts but in male hosts it inhibited enzyme activity of sarcoma 180 cells and activated activity in carcinoma cells. The effect of all 3 hormones on acid phosphatase activity was activation. With adenosine triphosphatase, estrogen stimulated the activity in both types of tumor in both sexes. Progesterone stimulated cells from male hosts with little or no effect on cells from female hosts. This enzyme was resistant to testosterone. Succinate dehydrogenase activity under similar conditions was different. Estrogen reduced this activity and progesterone produced some inhibition of activity. Testosterone inhibited the sarcoma cells but had no effect on carcinoma cells of either sex. Others have shown that sex hormones affect the enzyme activities beyond the target tissues, particularly in the liver, kidney, and pancreas. Different responses of the enzymes seemed to depend on the endogenous hormonal status of the mice.
